Polisot liegt etwa 35 Kilometer südöstlich von Troyes an der Seine. Nachbargemeinden von Polisot sind Bar-sur-Seine im Norden, Celles-sur-Ource im Osten, Polisy im Süden, Arrelles im Südwesten sowie Villemorien im Westen.
