Lerné liegt etwa zehn Kilometer westsüdwestlich von Chinon. Umgeben wird Lerné von den Nachbargemeinden Saint-Germain-sur-Vienne im Norden, Thizay im Norden und Nordosten, Seuilly im Osten, Vézières im Süden, Bournand im Süden und Südwesten, Roiffé im Westen sowie Couziers im Nordwesten.
