Papst Pius XII. errichtete das Bistum Trujillo am 4. Juni 1957 mit der Apostolischen Konstitution In maximis officii[1] aus Gebietsabtretungen des Erzbistums Mérida, dem es als Suffragandiözese unterstellt wurde.
