Trélou-sur-Marne liegt etwa 80 Kilometer ostnordöstlich von Paris an der Marne, die die Gemeinde im Süden begrenzt. Umgeben wird Trélou-sur-Marne von den Nachbargemeinden Courmont im Norden, Champvoisy im Nordosten, Vincelles im Osten, Dormans im Süden und Südosten, Courthiézy im Süden, Reuilly-Sauvigny im Südwesten, Barzy-sur-Marne im Westen sowie Le Charmel im Nordwesten.
